    A comparative spectroscopic and kinetic study of photoexcitations in detergent-isolated and membrane-embedded LH2 light-harvesting complexes

    AbstractIntegral membrane proteins constitute more than third of the total number of proteins present in organisms. Solubilization with mild detergents is a common technique to study the structure, dynamics, and catalytic activity of these proteins in purified form. However beneficial the use of detergents may be for protein extraction, the membrane proteins are often denatured by detergent solubilization as a result of native lipid membrane interactions having been modified. Versatile investigations of the properties of membrane-embedded and detergent-isolated proteins are, therefore, required to evaluate the consequences of the solubilization procedure. Herein, the spectroscopic and kinetic fingerprints have been established that distinguish excitons in individual detergent-solubilized LH2 light-harvesting pigment–protein complexes from them in the membrane-embedded complexes of purple photosynthetic bacteria Rhodobacter sphaeroides. A wide arsenal of spectroscopic techniques in visible optical range that include conventional broadband absorption–fluorescence, fluorescence anisotropy excitation, spectrally selective hole burning and fluorescence line-narrowing, and transient absorption–fluorescence have been applied over broad temperature range between physiological and liquid He temperatures. Significant changes in energetics and dynamics of the antenna excitons upon self-assembly of the proteins into intracytoplasmic membranes are observed, analyzed, and discussed.     Food choices, physical activity and metabolic health in obese patients

    The aim of the current study was to analyse the food choices and physical activity of obese adult Estonian persons, and associations with the prevalence of metabolic syndrome. The study was carried out on 76 patients aged over 35 years whose body mass index was ≄30 kg/m2. The subjects were recruited through family physicians. The subjects’ consumption of three food groups (fruit, vegetables, whole-grain products) and physical activity based on the IPAQ questionnaire was compared with the prevalence of metabolic syndrome based on five indicators (waist circumference, triglycerides, HDL- cholesterol and fasting plasma glucose, blood pressure). The prevalence of metabolic syndrome was found to be 50%. The results of the study did not show statistically significant correlations between prevalence of metabolic syndrome and age or gender. Neither were there any significant age or gender differences in the subjects’ nutritional and activity behaviour. Comparison of the nutritional behaviour of persons with and without the metabolic syndrome showed that daily consumers of fruit had a 4.48 times lower risk of metabolic syndrome than those who ate fruit more seldom. No statistically significant correlation was found between physical activity and prevalence of metabolic syndrome. Based on the current study, the daily consumption of fruit can be an essential protective factor against metabolic syndrome in obese patients and provides a simple recommendation physicians can give their patients to follow

    In situ high-resolution structure of the baseplate antenna complex in <i>Chlorobaculum tepidum</i>

    Photosynthetic antenna systems enable organisms harvesting light and transfer the energy to the photosynthetic reaction centre, where the conversion to chemical energy takes place. One of the most complex antenna systems, the chlorosome, found in the photosynthetic green sulfur bacterium Chlorobaculum (Cba.) tepidum contains a baseplate, which is a scaffolding super-structure, formed by the protein CsmA and bacteriochlorophyll a. Here we present the first high-resolution structure of the CsmA baseplate using intact fully functional, light-harvesting organelles from Cba. tepidum, following a hybrid approach combining five complementary methods: solid-state NMR spectroscopy, cryo-electron microscopy, isotropic and anisotropic circular dichroism and linear dichroism. The structure calculation was facilitated through development of new software, GASyCS for efficient geometry optimization of highly symmetric oligomeric structures. We show that the baseplate is composed of rods of repeated dimers of the strongly amphipathic CsmA with pigments sandwiched within the dimer at the hydrophobic side of the helix

    Family doctors' knowledge and self-reported care of type 2 diabetes patients in comparison to the clinical practice guideline: cross-sectional study

    BACKGROUND: It is widely believed that providing doctors with guidelines will lead to more effective clinical practice and better patient care. However, different studies have shown contradictory results in quality improvement as a result of guideline implementation. The aim of this study was to compare family doctors' knowledge and self-reported care of type 2 diabetes patients with recommendation standards of the clinical practice guideline. METHODS: In April 2003 a survey was conducted among family doctors in Estonia. The structured questionnaire focused on the knowledge and self-reported behavior of doctors regarding the guideline of type 2 diabetes. The demographic and professional data of the respondents was also provided. RESULTS: Of the 354 questionnaires distributed, 163 were returned for a response rate of 46%. Seventy-six percent of the responded doctors stated that they had a copy of the guideline available while 24% reported that they did not. Eighty-three percent of the doctors considered it applicable and 79% reported using it in daily practice. The doctors tended to start treatment with medications and were satisfied with treatment outcomes at higher fasting blood glucose levels than the levels recommended in the guideline. Doctors' self-reported performance of the tests and examinations named in the guideline, which should be performed within a certain time limit, varied from overuse to underuse. Blood pressure, serum creatinine, eye examination and checking patients' ability to manage their diabetes were the best-followed items while glycosylated hemoglobin and weight reduction were the most poorly followed. Doctors' behavior was not related to the fact of whether they had the guideline available, whether they considered it applicable, or whether they actually used it. CONCLUSION: Doctors' knowledge and self-reported behavior in patient follow-up of type 2 diabetes is very variable and is not related to the reported availability or usage of the guideline. Practice guidelines may be a useful source of information but they should not be overestimated

    Detecting ship wakes for the study of coastal processes

    Wakes from contemporary vessels may affect, and in some places dominate, coastal processes in the vicinity of major shipping lanes. The analysis of the properties and impact of wakes has generally been restricted to wakes that can be visually observed in raw data. In this work, spectral analysis of the time series of single-point measurements of water surface elevation from Tallinn Bay is used to highlight the structure of ship wakes using a Short Time Fourier Transform. This method makes it possible to determine the speed and distance of a vessel from the measurement site. Wakes are detected using an algorithm based on Gabor multipliers. The results are compared with vessel passages retrieved from the Automatic Identification System (AIS) data. The algorithm detects the majority of ship wakes that can be visually recognized in spectrograms and misses only those with low signal to noise ratio or those in close proximity to another vessel wake. The calculated speed and distance are consistent with the AIS data except for high-speed vessels sailing at ≄30 knots. The results indicate that by using these techniques the detection of vessel wakes from a single-point wave record is achievable under favorable weather conditions. The methods provide an option for mitigation of the impact of ship wakes in semi-enclosed water bodies
